Urinary Citrate

Method:Enzymatic, Manual

Principle:
Citrate  Lyase  catalyzes conversion of citrate to oxaloacetate and acetate. Same oxaloacetate may be decomposed to pyruvate by oxaloacetate decarboxylase which possibly contaminates the citrate lyase preparation . Both α-ketoacids react with phenydrazine at acidic pH and produce phenylhydrazone which absorbes the light at 340 nm. The absorbance is directly proportional to citrate concentration.

Specimens:
Twenty four-hour urine sample collected in a bottles containing 10 ml concentrated HCl can be used.
