Method:Berthelot Method, Manual

Principle:
Urea is converted to ammonium ion and carbondioxide by urease. The ammonium ion produces a complex with salicylate and hypochlorite ions which absorbs the light at 578 nm. The absorbance is proportional to the urea concentration.

Specimens:
    Either serum or plasma can be used for analysis. If plasma is to be used anticoagulants containing ammonium or fluoride ions must be avoided. This kit can also be used to determine the concentration of urea in urine.

Reagent 1:  Buffer Solution (pH 7.2)           2 x 50 ml
Reagent 2: Enzyme Solution                       1 ml
Reagent 3: Sodium-hypochlorite                 2 x 50 ml
Reagent 4: Standard Solution (50 mg/dl)     5 ml
