Method:Colorimetric, End-point, Manual/Automated

Principle:
The biuret method is used to determine total protein levels. The reaction between proteins and cupric ions forms a violet-colored complex. The intensity of the color is measured colorimetrically (546nm) and is directly proportional to the concentration of proteins.

Specimens:
Serum is the sample of choice since the fibrinogen present in plasma is a protein and interferes with the analysis. Hemolyzed serum should not be used.
